Product Specific Information

The 30-H12 monoclonal antibody reacts with mouse CD90.2 (Thy-1.2), a GPI-linked membrane molecule expressed by mouse thymocytes, mature T cells, and neurons in CD90.2-expressing strains (BALB/c, CBA, C3H, C57BL/6, C58/J, SJL, etc.). Cells from CD90.1-expressing strains (PL, AKR) do not stain with 30-H12. CD90 is involved in T cell adhesion and signal transduction.

Applications Tested: Mouse splenocytes (≤ 0.5 µg/test, 100 µL final volume, 10⁵–10⁸ cells/test)

Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended detection filter: 780/60 bandpass. Ensure instrument compatibility.

Compensation Notes:

• Higher compensation values may occur in violet 450/50 channel when using UltraComp eBeads (#01-2222-42) compared to stained cells.
• Compensation with cells is recommended.
• Similar observations with AbC Total Antibody Compensation Beads (#A10497).

Staining Buffer Recommendation:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (#SB-4401) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (#00-8222-49)
• 1-step Fix/Lyse Solution (#00-5333-57)

Target Information

CD90.2 (Thy-1) is a GPI-linked glycoprotein of the Immunoglobulin superfamily. Expressed in murine T cells, thymocytes, neural cells, Kupffer cells, and fibroblasts. Plays roles in cell-cell and cell-ligand interactions during synaptogenesis. CD90.2 (18 kDa) is found on hematopoietic cells, neurons, connective tissue, fibroblasts, stromal cells, and tumor endothelial cells. In mice, expressed on all thymocytes and peripheral T lymphocytes.
