Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, and binds glycosylated, fucosylated, and sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCam-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules of peripheral lymphoid tissues.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse splenocytes at ≤0.03 µg/test (100 µL final volume). Optimal performance requires titration.
• Fluorophore Properties: Super Bright 780 is excited at 405 nm and emits at 780 nm; use a 780/60 bandpass filter.
• Staining Recommendation: Use Super Bright Complete Staining Buffer (SB-4401) when using multiple Super Bright conjugates.
• Compensation Notes: When using UltraComp eBeads (01-2222-42) or AbC Total Antibody Compensation beads (A10497), compensation values may vary; use cells for accurate compensation.
• Light Sensitivity: Protect from light to prevent photo-induced oxidation.
• Fixation: Samples can be stored in IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333) for up to 3 days at 4°C in the dark.
• Laser Compatibility: Violet laser (405 nm).

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor consisting of:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Transmembrane and cytoplasmic regions

It is expressed on all leukocytes except a subset of memory T cells, and plays a role in leukocyte adhesion and migration.
