Target Information
RIP1 is a serine-threonine kinase that mediates inflammatory and programmed necrosis signaling following activation of death receptors, PRRs, and DNA damage. Upon TNFR1 activation by TNF-alpha cytokines, TRADD and TRAF2 are recruited to the receptor. RIP1 phosphorylates DAB2IP at Ser-728 in a TNF-alpha-dependent manner, activating the MAP3K5-JNK apoptotic cascade.
Ubiquitination by TRAF2 via Lys-63-linked chains enhances communication with downstream signal transducers in the MAPK and NF-kappa-B pathways, leading to activation of inflammatory genes. Polyubiquitinated RIP1 binds to IKBKG/NEMO, a key step for NF-kappa-B activation. Interaction with other RHIM-containing adapters initiates gene activation and cell death. RIPK1 and RIPK3 association forms a necrosis-inducing complex.
