Product Specific Information

The RMP1-30 antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. It is expressed mainly on activated T and B lymphocytes.

Two B7 family members, PD-L1 (B7-H1) and PD-L2 (B7-DC), act as PD-1 ligands. These ligands show overlapping functions and are expressed on normal tissues and upregulated on activated antigen-presenting cells. RMP1-30 does not block the binding of either B7-H1-Ig or B7-DC-Ig to PD-1 transfectants.

Applications Tested: Tested on Con A-stimulated mouse splenocytes at ≤0.25 µg/test.
A test is defined as the amount of antibody that stains a cell sample in 100 µL volume. Cell count range: 10⁵–10⁸ cells/test.
Antibody titration is recommended for optimal performance.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

T-cell activation is self-limited through receptors such as PD-1 (PDCD-1) that deliver inhibitory signals upon ligand binding. PD-1 binds PD-L1 and PD-L2, and this interaction suppresses immune activation in the absence of danger signals. PD-1 deficiency leads to hyperactivated immune responses and autoimmune diseases. Although PD-1 has a predicted molecular weight of 55 kDa, it often migrates at a higher molecular weight in SDS-PAGE.
