Product Specific Information

The LN3 monoclonal antibody reacts with the human major histocompatibility complex (MHC) class II molecule, HLA-DR. HLA-DR is expressed on antigen-presenting cells (APCs) such as B cells, monocytes, macrophages, dendritic cells, and activated T cells. It is a heterodimeric transmembrane protein composed of alpha and beta subunits and plays a key role in peptide presentation to CD4⁺ T lymphocytes.

  • Applications Tested: Tested by flow cytometric analysis of normal human peripheral blood cells.
    Recommended use ≤ 0.0075 µg per test (100 µL final volume).
    Optimal titration is advised for best assay performance.
  • Filtration: 0.2 µm post-manufacturing filtered.

Target Information

HLA-DR is a transmembrane glycoprotein consisting of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB). The alpha chain gene includes 5 exons encoding leader peptide, extracellular domains, transmembrane domain, and cytoplasmic tail. DRA serves as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5. The beta chain contains the polymorphisms determining peptide-binding specificity.

HLA-DR is mainly expressed on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
MHC class II molecules (DR, DQ, DP) bind processed peptides and present them to T-helper cells, initiating immune responses.

HLA and MHC antibodies are critical in Immunopeptidomics, aiding in the identification and characterization of neoantigens via LC-MS/MS.
