Target Information
TNF alpha is a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as a multimer of two, three, or five noncovalently linked units, showing a single 17 kDa band following SDS-PAGE under non-reducing conditions. Knockout studies in mice indicate a neuroprotective function, and TNF alpha has been observed to induce tumor necrosis in tumor-bearing mice. It also plays key roles in immune responses to bacterial, viral, parasitic, and fungal infections, as well as in tumor necrosis. TNF alpha can cause cytolysis or cytostasis of certain transformed cells, synergistically with interferon-gamma. It has been implicated in autoimmune diseases, insulin resistance, and cancer.
