Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers of two to five noncovalently linked units and shows a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies in mice suggest neuroprotective functions, and TNF alpha induces tumor necrosis in tumor-bearing mice. It plays roles in immune responses to bacterial, viral, parasitic, and fungal infections and contributes to tumor cell cytolysis, synergistic with interferon-gamma. TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
