Product Specific Information
Reconstitution:
Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/mL or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.
Target Information
CD22 (BL-CAM) is a type 1 integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation alongside CD19 antigen. Membrane expression is limited to mature B cells (CD22+) and plasma cells (CD22−), making it useful in phenotyping mature leukemias. CD22 is strongly expressed in hairy cell leukemia and binds preferentially to α2,6-linked sialic acid. This recognition site can be masked by cis interactions on the same cell surface. Ligand-induced tyrosine phosphorylation regulates B-cell receptor signaling, acting both positively via Src family kinases and negatively via phosphatase recruitment. CD22 is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
