Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor (~90 kDa) has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR is a ligand-activated transcription factor that binds active testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

The AR signaling pathway is essential for the development and function of male reproductive organs (prostate, epididymis) and also plays roles in muscle, hair follicles, and brain. AR is a phosphoprotein and regulates MAP kinase pathways. Dysregulation of AR signaling is associated with prostate cancer, Kennedy’s disease, male infertility, and complete androgen insensitivity syndrome (CAIS).
