Target Information
KIT (c-KIT)는 MGF(mast cell growth factor, stem cell factor)의 type 3 transmembrane receptor로서, 세포 성장과 활성 조절에 관여하는 proto-oncogene입니다. KIT 유전자는 고양이 육종 바이러스 유래 v-kit의 세포 상동체로 처음 확인되었습니다. KIT와 그 리간드는 조혈 및 비조혈 세포의 성장과 활성화를 조절하며, KIT 변이는 위장관 기질종양(GIST), 비만세포 질환, 급성 골수성 백혈병, piebaldism 등과 관련이 있습니다. 특히 D816V 활성 루프 변이는 비정상적인 KIT 수용체 활성화를 유발하여 치료 전략의 주요 표적이 됩니다. c-Kit은 조혈 줄기세포, 생식세포, 비만세포, 위장관의 Cajal 세포에서 발현되며, SCF(stem cell factor) 결합 후 수용체 이합체화 및 자가 인산화가 일어나 다양한 신호전달 경로(Grb2/Ras/ERK, PI3K, PLC-γ, JAK/STAT)를 활성화합니다. c-Kit 수용체 활성은 조혈, 줄기세포 유지, 생식세포 형성에 필수적입니다.
