Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding allows ER to regulate gene transcription through coactivator or corepressor proteins.
There are two forms of estrogen receptor, alpha and beta, encoded by separate genes (ESR1 and ESR2). Alternative RNA splicing generates at least four alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa).
Estrogen receptors are widely expressed and play key roles in sexual development, reproductive function, bone physiology, and pathological processes such as breast cancer, endometrial cancer, and osteoporosis.
