Product Specific Information
The SK7 monoclonal antibody reacts with human and chimpanzee CD3ε, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (γ and δ), the ε chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. SK7 and UCHT1 clones cross-block binding, suggesting recognition of overlapping epitopes, whereas OKT3 and SK7 recognize distinct epitopes.

Target Information
The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. T cell activation through the TCR involves CD3γ, CD3δ, CD3ε, and CD3ζ subunits, which are structurally related members of the immunoglobulin superfamily encoded on chromosome 11. CD3 subunits associate with cytoplasmic signaling molecules via double tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Reduced T lymphocyte percentages may occur in certain autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
