Target Information
Apoptosis plays a major role in organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process is linked to diseases such as cancer. Members of the Bcl-2 family regulate apoptosis through conserved Bcl-2 homology (BH) domains.
Bcl-rambo, a novel and widely expressed member, localizes to mitochondria and induces apoptosis, which can be blocked by co-expression of inhibitor of apoptosis proteins (IAPs) such as XIAP, cIAP1, and cIAP2.
Unlike Bcl-2, Bcl-rambo contains a unique 250 amino acid insertion before its C-terminal membrane anchor. This region alone can induce apoptosis more efficiently than the BH domains, suggesting a role in other pro-apoptotic pathways.
