Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) present on many stem cell populations. It serves as a stem cell marker, with reversible expression in human hematopoietic stem cells. CD34 may function as a surface receptor involved in receptor-mediated endocytosis and regulation of adhesion, differentiation, and proliferation of hematopoietic stem and progenitor cells.
CD34 expression indicates a specific hematopoietic developmental state, influencing adhesion properties and differentiation potential both in vitro and in vivo. It may act as an adhesion molecule mediating stem cell attachment to the bone marrow extracellular matrix or stromal cells, and as a scaffold for lineage-specific glycans, facilitating binding to lectins expressed by stromal or marrow components.
Additionally, CD34 may present carbohydrate ligands to selectins. The intracellular domain is phosphorylated by activated protein kinase C, suggesting a role in signal transduction. CD34 dysfunction is associated with diseases such as dermatofibrosarcoma and neurofibroma.
