Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a stem cell marker and may function as a surface receptor involved in receptor-mediated endocytosis, adhesion, differentiation, and proliferation of hematopoietic stem and progenitor cells. CD34 expression represents a specific developmental stage of hematopoietic cells with altered adhesion properties.
CD34 may mediate attachment of stem cells to the bone marrow extracellular matrix or stromal cells and act as a scaffold for glycans that interact with lectins on stromal or marrow components. It is also implicated in presenting carbohydrate ligands to selectins. The intracellular domain can be phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
