Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
p27 Kip1 functions as a negative regulator of G1 phase progression and may act as a mediator of TGFβ-induced G1 arrest. It is a potent inhibitor of cyclin E- and cyclin A-CDK2 complexes and interacts with cyclin D-CDK4 complexes to modulate their activation. Depending on its phosphorylation state, p27 Kip1 can act as an inhibitor or activator of cyclin D-CDK4 complexes. The protein has an approximate molecular weight of 27 kDa, and its degradation via CDK-dependent phosphorylation and ubiquitination by SCF complexes is essential for the transition from quiescence to proliferation.
