Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

p27 Kip1 functions as a negative regulator of G1 phase progression and may mediate TGFβ-induced G1 arrest. It is a key inhibitor of cyclin E- and cyclin A-CDK2 complexes and interacts with cyclin D-CDK4 complexes, influencing their assembly and activation. Depending on phosphorylation state and stoichiometry, p27 Kip1 can act as either an inhibitor or activator of cyclin D-CDK4 complexes.
This 27 kDa protein undergoes CDK-dependent phosphorylation and ubiquitination via SCF complexes, enabling the transition from quiescence to proliferation.
