Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy. PR exists as two forms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B) due to alternative translation initiation sites. PR-B is transcriptionally active and responsible for gene activation related to endometrium maintenance, pregnancy, and inhibition of ovulation.

PR-A is identical to PR-B except for a 165 amino acid deletion at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity. In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, conformational changes enable dimerization and binding to progesterone response elements (PRE), resulting in activated transcription. Null mutations in the PGR gene lead to reproductive abnormalities.
