Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. Its main function is to promote tubulin polymerization and stabilize microtubules. The C-terminus binds axonal microtubules, while the N-terminus binds neural plasma membrane components, suggesting Tau functions as a linker between both structures. Axonal polarity is influenced by TAU/MAPT localization within the neuronal cell body domain defined by the centrosome.
Short Tau isoforms contribute to cytoskeletal plasticity, whereas longer isoforms aid in stabilization. Hyper-phosphorylated Tau is the major component of paired helical filaments (PHF), which form neurofibrillary lesions in Alzheimer’s disease (AD). This modification impairs Tau’s microtubule binding ability, leading to microtubule destabilization and neuronal degeneration.
Tau hyper-phosphorylation is mediated by kinases such as GSK-3β, PKA, cdk5, and casein kinase II. Abnormal Tau aggregation is observed in several neurodegenerative disorders, including Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
