Target Information
HLA-DRB5는 HLA class II beta chain paralogues에 속하며, alpha (DRA) 및 beta (DRB) 체인으로 구성된 이종이합체입니다. 이 분자는 항원제시세포(APC: B lymphocytes, dendritic cells, macrophages)에서 발현되며, 세포 외 단백질 유래 펩타이드를 제시하여 면역계에서 중심적인 역할을 합니다.
Beta 체인은 약 26–28 kDa이며, 6개의 엑손으로 구성되어 있습니다. Exon 1은 leader peptide, Exon 2와 3은 extracellular domain, Exon 4는 transmembrane domain, Exon 5는 cytoplasmic tail을 암호화합니다.
DR 분자 내에서 beta 체인은 펩타이드 결합 특이성을 결정하는 다형성을 포함합니다. 이러한 다형성은 골수 및 신장이식 시 정기적으로 타이핑됩니다. DRB1은 DRB3, DRB4, DRB5보다 약 5배 높은 수준으로 발현되며, DRB5의 존재는 DRB1의 대립유전자 변이와 연관되어 있습니다. 관련된 pseudogene으로는 DRB2, DRB6, DRB7, DRB8, DRB9가 있습니다.
HLA 및 MHC 항체는 면역펩티도믹스 연구에서 중요한 역할을 하며, 고성능 액체크로마토그래피와 tandem 질량분석을 결합하여 신항원(neoantigen)을 식별하고 특성화하는 데 사용됩니다.
