Target Information
Apoptosis is related to many diseases and developmental processes. The p53 tumor-suppressor protein induces apoptosis through transcriptional activation of several genes including p53R2, p53AIP1, and PUMA. A newly identified p53 target gene, Noxa, encodes a protein belonging to the BH3-only proapoptotic subfamily. Both Noxa and PUMA are transcriptional targets of p53 and BH3-only proteins.
X-ray irradiation increases p53-dependent Noxa mRNA and protein levels. When ectopically expressed, Noxa interacts with anti-apoptotic Bcl-2 family members, activating caspase-9. Like PUMA, Noxa localizes to mitochondria and induces apoptosis in response to p53. Noxa and PUMA may act as direct mediators of p53-induced apoptosis. Elevated levels of p53 and Noxa are associated with impaired tumor development.
