This 8CH monoclonal antibody reacts with human IL-8 (CXCL8), a pro-inflammatory CXC chemokine. It is synthesized as a 99 amino acid precursor protein that is processed into one of four isoforms, typically 72 or 77 amino acids in length.
IL-8(77) is secreted mainly by endothelial cells and is less potent in neutrophil activation compared to IL-8(72). During fetal development, IL-8(77) mediates angiogenesis rather than inflammation.
The predominant adult form, IL-8(72), is expressed by monocytes, macrophages, epithelial cells, and fibroblasts in response to inflammatory stimuli, stress, and hormones. It is crucial for neutrophil activation and recruitment and influences T cell migration.
Signaling occurs via CXCR1 and CXCR2 receptors. IL-8 expression is often elevated in tumors, contributing to angiogenesis and metastasis.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for use at 5 µL (0.015 µg)/test. One test stains cells in a 100 µL final volume, typically 10⁵–10⁸ cells/test.

Interleukin 8 (IL-8, CXCL8) is a 72 amino acid pro-inflammatory chemokine of the CXC family, binding to IL-8RA and IL-8RB receptors.
It acts as a chemoattractant and angiogenic factor, induced by inflammatory stimuli in cells such as macrophages and endothelial cells.
In endothelial cells, IL-8 is stored in Weibel-Palade bodies.
IL-8 signals through CXCR1 and CXCR2 and contains the ELR motif (Glu-Leu-Arg).
It was previously known as NAP-1, GCP-1, MONAP, and 3-10C.
Located on chromosome 4q, IL-8 is part of a chemokine gene cluster.
It is implicated in tumor angiogenesis, growth, and bronchiolitis pathogenesis.
