Target Information
Peroxisome proliferators are non-genotoxic carcinogens that exert their effects through interaction with members of the nuclear hormone receptor family termed peroxisome proliferator-activated receptors (PPARs). These nuclear hormone receptors are ligand-dependent intracellular proteins that regulate transcription of specific genes by binding to DNA sequences upon activation by appropriate ligands. PPARs are activated by compounds such as clofibric acid, nafenopin, and WY-14,643, as well as by some fatty acids. They can induce transcription of acyl coenzyme A oxidase and cytochrome P450 (CYP450) A6 through specific response elements. PPARs also heterodimerize with retinoid X receptor (RXR) alpha, similar to other nuclear hormone receptors.
