Preparation:
Produced from sera of goats immunized with highly pure Recombinant Murine TNF-alpha. Anti-Murine TNF-alpha-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect Murine TNF-alpha by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL is required.
This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Murine TNF-alpha (500-P64G) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Murine TNF-alpha.

Western Blot:
To detect Murine TNF-alpha by Western Blot, use at 0.1–0.2 µg/mL.
With compatible secondary reagents, detection limit for Recombinant Murine TNF-alpha is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.

It exists as multimers (dimers, trimers, or pentamers) but shows a single 17 kDa band on SDS-PAGE under non-reducing conditions. Knockout studies in mice suggest a neuroprotective function. TNF alpha induces tumor necrosis when injected into tumor-bearing mice and plays roles in immune responses to bacterial, viral, parasitic, and fungal infections. It also contributes to cytolysis or cytostasis of transformed cells, synergistically with interferon-gamma. TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
