Target Information

CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, a subpopulation of T lymphocytes, and follicular dendritic cells. CD20 expression is synchronous with surface IgM and regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 associates with lipid rafts, with association intensity depending on extracellular triggers and BCR aggregation. After receptor ligation, CD20 and BCR colocalize and then dissociate before BCR endocytosis, while CD20 remains at the cell surface.
CD20 is a useful target for antibody-mediated B-cell depletion therapy, as it is highly expressed on most B-cell malignancies and remains on the plasma membrane without internalization or shedding after monoclonal antibody treatment.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
