Description:
The 6C4 monoclonal antibody reacts with human CD32 (Fc gammaRII), recognizing isoforms FcγRIIA and FcγRIIB. These 40 kDa polymorphic transmembrane glycoproteins are expressed on B cells, granulocytes, monocytes, macrophages, and platelets, as well as NK cells. CD32 mediates interaction between FcγRII-expressing cells and opsonized antigens or IgG immune complexes, playing roles in inflammation and autoimmune disease. Clone 6C4 has been reported to inhibit Ig binding in a rosette blocking assay.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per 100 µL cell sample (10⁵–10⁸ cells/test).

Light Sensitivity: Protect from light due to photo-induced oxidation of tandem dye.
Fixation: Samples may be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
Excitation/Emission: 488–561 nm / 775 nm (Blue, Green, Yellow-Green Laser).
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD32 encodes an Fc receptor found on phagocytic cells (macrophages, neutrophils) that mediates phagocytosis and clearance of immune complexes. Multiple transcript variants are produced by alternative splicing.
