Product Specific Information
The P67.6 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein belonging to the Siglec (sialic acid-binding Ig superfamily lectin) family. CD33 is expressed in the hematopoietic compartment, including monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Applications Tested:
- Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells
- Recommended usage: 5 µL (0.125 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test)
Handling Notes:
- Light sensitivity: Protect from light due to photo-induced oxidation sensitivity
- Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark
- Excitation/Emission: 488–561 nm / 775 nm
- Compatible Lasers: Blue, Green, Yellow-Green Lasers
Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases and regulate receptor signaling.
CD33 is expressed on cells of the myelomonocytic lineage, including granulocyte and macrophage precursors, and is a therapeutic target for acute myeloid leukemia (AML). It is also a useful marker for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
