Target Information
Low density lipoprotein-related protein 2 (LRP2), also known as megalin, is a multi-ligand endocytic receptor expressed mainly in absorptive epithelial tissues such as the kidney. It possesses a large extracellular domain, a single transmembrane domain, and a short cytoplasmic tail. LRP2 binds various macromolecules, including albumin, apolipoproteins B and E, and lipoprotein lipase, and is essential for reuptake of ligands like lipoproteins, sterols, vitamin-binding proteins, and hormones.
It also plays a role in cell signaling, interacting with parathyroid hormones and sonic hedgehog extracellularly, and MAP kinase scaffold proteins and JNK interacting proteins intracellularly. Recycling of LRP2 is regulated by phosphorylation of its cytoplasmic domain.
Mutations in LRP2 cause Donnai-Barrow syndrome (DBS) and facio-oculoacoustico-renal syndrome (FOAR).
