Semaphorins constitute the largest family of axon guidance cues described to date. They are divided into eight classes, with classes 3–7 found in vertebrates.
- Class 3 Semaphorins: Secreted proteins
- Classes 4–6: Transmembrane proteins
- Class 7: Membrane-associated via GPI linkage
Semaphorins possess a conserved ~400 amino acid sema domain and are known as collapsing factors and mediators of axon repulsion, while also functioning as context-dependent chemoattractants.
They play roles in cardiovascular development and immune cell antigen presentation.
Receptors mediating semaphorin signaling include Neuropilin and Plexin family proteins.
