• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature (RT).
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at RT for 20 min.

Target Information

CD63 (LAMP-3, lysosome-associated membrane protein-3)는 tetraspanin 계열의 당단백질로, 다양한 세포의 후기 엔도솜, 리소좀 및 분비 소포에 존재합니다. 세포 활성화 후에는 세포막에서도 발견되며, basophil 활성화 마커로 널리 사용됩니다.
CD63은 integrin과 상호작용하여 식세포 작용 및 세포 이동에 영향을 미치며, H/K-ATPase 및 ROMK1 채널 조절에도 관여합니다. 또한 T 세포 공동자극 분자로 작용하며, 암의 초기 단계 예후 예측에도 활용될 수 있습니다.
활성화된 혈소판, 단핵구, 대식세포에서 발현되며, 과립구, B, T 세포에서는 약하게 발현됩니다. CD63은 흑색종 관련 항원 ME491 및 혈소판 항원 PTLGP40과 동일합니다.
CD63 기능 이상은 흑색종 및 Hermansky-Pudlak 증후군과 관련이 있습니다.
