- Immunohistochemistry (PFA fixed):
- Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues, heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
CD63 (LAMP-3, lysosome-associated membrane protein-3)은 tetraspanin 계열의 당단백질로, 다양한 세포의 후기 엔도좀, 리소좀 및 분비 소포에 존재합니다. 세포 활성화 후에는 세포막에서도 발견됩니다.
CD63은 basophil 활성화 마커로 널리 사용되며, integrin과 상호작용하여 식세포작용 및 세포 이동에 영향을 미칩니다. 또한 H/K-ATPase 및 ROMK1 채널의 조절, T세포 공동자극에도 관여합니다.
CD63 발현은 암의 예후 예측에 활용될 수 있으며, 활성화된 혈소판, 단핵구, 대식세포에서 발현되고, 과립구, B세포, T세포에서는 약하게 발현됩니다.
CD63은 melanoma-associated antigen ME491 및 platelet antigen PTLGP40과 동일 단백질입니다.
CD63 이상은 흑색종(melanoma) 및 Hermansky-Pudlak 증후군과 관련이 있습니다.
