Product Specific Information

The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven transmembrane G-protein-linked chemokine receptor binding CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 plays a role in CD4 T cell responses to grafts, and knockout mice show compromised allograft rejection. Expression is found on NK cells, subsets of T lymphocytes, Tregs, and Th1-polarized cells.

This antibody affects chemotaxis in response to ligand; ligand presence eliminates staining. In vivo addition delays cardiac and pancreatic islet allograft rejection.

Super Bright 600 is a tandem dye excited by a 405 nm violet laser and emits at 600 nm. Use a 610/20 bandpass filter. Ensure the instrument supports this fluorochrome.

When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.

Target Information

This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC, members of the CXC chemokine subfamily. Chemokine binding induces leukocyte trafficking, integrin activation, cytoskeletal changes, and chemotaxis. Inhibition by Bordetella pertussis toxin indicates coupling to Gi-class G proteins.

Expression is prominent in effector/memory T cells and inflamed tissues. IP10, Mig, and I-TAC production in inflammatory lesions suggests involvement in immune cell recruitment. Thus, this receptor is a potential target for small-molecule antagonists for inflammatory disease treatment. Multiple transcript variants exist.
