Target Information

NUAK1 (ARK5) is a serine/threonine kinase involved in multiple cellular processes. Overexpression is linked to tumor invasion and metastasis, particularly in colorectal neoplasms. It regulates cellular senescence and ploidy via phosphorylation of LATS1 at Ser-464, influencing its stability. NUAK1 also controls cell adhesion through modulation of myosin protein phosphatase 1 (PP1) activity by phosphorylating PPP1R12A, leading to reduced dephosphorylation of MLC2. It may participate in DNA damage response by phosphorylating p53 at Ser-15 and Ser-392 and acting in CDKN1A promoter activation. NUAK1 can promote tumor malignancy under AKT1 regulation and suppress Fas-induced apoptosis via CASP6 phosphorylation. It also contributes to UV-induced DNA damage response and CDKN1A degradation for optimal DNA repair.
