The LG.3A10 monoclonal antibody reacts with mouse CD27, a lymphocyte-specific member of the TNFR superfamily.
CD27 is expressed by nearly all mature T cells and by a subset of B cells, mainly memory B cells. In mice, CD27 is found on most thymocytes except CD46-CD8- precursors. CD27 binds to CD70, playing a key role in T cell–B cell interactions and effector cell maturation.

When LG.3A10 is crosslinked on purified T cells, proliferation increases; without crosslinking, it inhibits CD27–CD70 interaction.

Applications Reported: Flow cytometric analysis, immunoprecipitation, and immunohistochemical staining.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Cell number should be optimized (10⁵–10⁸ cells/test). Careful titration is recommended for optimal performance.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily, including CD40 and CD30. These receptors regulate cell proliferation and death.
CD27 and its ligand CD70 are mainly expressed in lymphocytes. High CD27 expression depends on antigen receptor ligation, while CD70 requires co-stimulatory signals.

CD27 exists as a disulfide-linked homodimer on mature thymocytes, peripheral T cells, and a subset of B cells. Activation via TCR-CD3 increases CD27 expression and releases soluble CD27 (sCD27), a 28–32 kDa form detected in plasma, urine, or spinal fluid. sCD27 serves as a prognostic marker for B cell malignancies.

RgpA, a cysteine proteinase, degrades CD27 and counteracts CD27-mediated activation. CD27-binding protein (SIVA), a proapoptotic molecule, interacts with CD27 and contributes to apoptosis signaling.
