Product Specific Information

Description:
The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ peripheral T lymphocytes as well as B cells. CD45RA+ T cell populations are mainly naive/virgin, allowing use of HI100 mAb as a phenotypic marker to discriminate T cell subsets.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended 5 µL (0.25 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test (empirical determination required).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It regulates Src-family kinases, essential for lymphocyte development and antigen signaling.
CD45 isoforms arise from alternative splicing and differ in extracellular domains but share identical transmembrane and cytoplasmic regions. Expression depends on cell type and physiological state. CD45 also promotes cell survival via integrin-mediated pathways and participates in apoptosis-related DNA fragmentation.
Defects in CD45 function are linked to severe combined immunodeficiency and Hepatitis C. CD45 is absent in mature red cells and non-hematopoietic tissues.
