Product Specific Information

Description:
The 104 monoclonal antibody reacts with mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.2-expressing mouse strains such as C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR. CD45.2 is expressed by all leukocytes in these strains.

Applications Reported:
Validated for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.5 µg per test (100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Optimal concentration should be titrated for specific assays.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and compensation efficiency.

Target Information

CD45.2 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising about 10% of surface proteins in lymphocytes. It regulates Src-family kinases, influences lymphocyte development and antigen signaling, and contributes to integrin-mediated signal transduction and apoptosis regulation. CD45RA is an isoform with restricted expression among lymphoid subtypes.
