Product Specific Information

Description:
The UCHL1 monoclonal antibody reacts with human CD45RO, a 180 kDa isoform of CD45. CD45RO is expressed by most thymocytes, activated memory T cells, granulocytes, and monocytes. CD22 is a ligand for CD45RO. Expression of CD45RO and CD45RA is commonly used to distinguish subsets of peripheral T cells.

Applications Reported:
This UCHL1 antibody is suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test (100 µL total volume). Cell number: 10⁵–10⁸ cells/test (empirically determined).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD45RO (UCHL1, ubiquitin carboxyl terminal esterase L1) is the shortest isoform of the receptor-type protein tyrosine phosphatase CD45. CD45 is essential for lymphocyte development and antigen signaling, regulating Src-family kinases, promoting cell survival via integrin-mediated pathways, and participating in apoptosis-related DNA fragmentation. CD45 isoforms share identical transmembrane and cytoplasmic domains but differ in extracellular regions, affecting localization and cell-type expression. CD45RO is expressed on macrophages, CD8⁺ T cells, activated T cells, and myeloma cells.
