Product Specific Information

The RMT3-23 monoclonal antibody reacts with mouse CD366 (TIM3), a Th1-specific cell surface protein. It recognizes CD366 expressed in both BALB/c and C57BL/6 mouse strains. CD366 is a type I transmembrane protein containing an immunoglobulin and mucin-like domain extracellularly, and a tyrosine phosphorylation motif intracellularly.

CD366 is selectively expressed by differentiated CD4⁺ Th1 and CD8⁺ Tc1 cells, but absent on Th2/Tc2 cells and other hematopoietic cell types such as naïve T cells, B cells, macrophages, and dendritic cells. Expression increases at late stages of Th1 differentiation, suggesting roles in effector function rather than differentiation.

In experimental autoimmune encephalomyelitis (EAE), CD366 is expressed on most CD4⁺ and CD8⁺ T cells in the CNS at disease onset, while peripheral CD4⁺ cells show minimal expression.

Functional activity: RMT3-23 blocks DC recognition of apoptotic cells and induces autoantibody production.

Applications Tested: Tested on mouse splenocytes at ≤ 0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration per assay.

Target Information

TIM3 (HAVCR2) is a 281-amino acid, type I Th1-specific cell surface glycoprotein expressed on terminally differentiated CD4⁺ Th1 and CD8⁺ Tc1 cells. It includes an IgV-like and mucin-like extracellular domain, and a conserved tyrosine phosphorylation motif intracellularly. TIM3 is involved in macrophage activation, autoimmune disease induction, and regulation of Th1-mediated immune responses, promoting immune tolerance. It has pathological relevance in EAE (Experimental Autoimmune Encephalomyelitis) and can enhance disease severity in mice.
