Target Information

Connexin 43 (Cx43) is a member of the gap junction protein family. Connexins assemble as hexamers and are transported to the plasma membrane to form hemichannels that connect adjacent cells, creating intercellular channels known as gap junctions. These structures are essential for cell-to-cell communication, development, and growth regulation.

Phosphorylation of Cx43 regulates gap junction assembly and function. Protein kinase C (PKC) phosphorylates Ser368, reducing intercellular communication, while Src kinase also modulates Cx43 phosphorylation. Connexin 43 is the major gap junction protein in the heart, crucial for synchronized cardiac contraction and embryonic development. Mutations in related genes can cause various inherited disorders, including Charcot-Marie-Tooth disease and heart malformations.
