Background
This gene encodes a member of the C1 family of peptidases. Alternative splicing results in multiple transcript variants, some of which encode preproproteins processed into multiple protein products, including Cathepsin B light and heavy chains that form a double-chain enzyme. This lysosomal cysteine protease exhibits both endopeptidase and exopeptidase activity, contributing to protein turnover. It also acts as amyloid precursor protein secretase, involved in APP proteolytic processing. Incomplete APP processing is implicated in Alzheimer’s disease. Overexpression has been linked to esophageal adenocarcinoma and other tumors.
