Cellular Localization Cytoplasm, Nucleus, Cell membrane. Translocates to the nucleus after activation by ILK1. Localization to the cell membrane occurs upon phosphorylation by TNK2, followed by further phosphorylation at Thr-308 and Ser-473, leading to activation. Colocalizes with WDFY2 in intracellular vesicles (PubMed:16792529).

Background

The serine-threonine protein kinase encoded by the AKT1 gene is catalytically inactive in serum-starved fibroblasts but becomes activated by platelet-derived growth factor. Activation occurs through phosphatidylinositol 3-kinase and is essential for growth factor-induced neuronal survival. AKT1 phosphorylates and inactivates apoptotic machinery components, suppressing apoptosis. Mutations in AKT1 are associated with Proteus syndrome. Multiple alternatively spliced transcript variants have been identified (RefSeq, Jul 2011).
