세포 내 위치

Cytoplasm and nucleus.
Cytoplasmic and nuclear in the absence of TGF-beta. Upon TGF-beta stimulation, migrates to the nucleus when complexed with SMAD4 (PubMed:9865696, PubMed:21145499).
Dephosphorylation by phosphatase PPM1A releases it from the SMAD2/SMAD4 complex, allowing export from the nucleus via interaction with RANBP1 (PubMed:16751101, PubMed:19289081).
Localized mainly to the nucleus in early embryo development, later expressed in the cytoplasm at blastocyst and epiblast stages (By similarity).

Background

The protein encoded by this gene belongs to the SMAD family, homologous to the Drosophila mothers against decapentaplegic (Mad) and C. elegans Sma genes.
SMAD proteins act as signal transducers and transcriptional modulators mediating multiple signaling pathways.
This protein mediates the TGF-beta signal and regulates cellular processes such as proliferation, apoptosis, and differentiation.
It is recruited to TGF-beta receptors via interaction with SARA (SMAD anchor for receptor activation).
Upon TGF-beta stimulation, it is phosphorylated by TGF-beta receptors, leading to dissociation from SARA and association with SMAD4, which is critical for nuclear translocation and subsequent signal transduction.
