Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor that, upon estrogen binding, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction regulates gene transcription through coactivator or corepressor proteins.
Two forms of estrogen receptors exist:
- Alpha (ESR1)
- Beta (ESR2)
Alternative RNA splicing produces at least four isoforms of estrogen receptor-alpha (Isoform 1: 66 kDa, Isoform 2: 53 kDa, Isoform 3: 47 kDa, Isoform 4: 35 kDa).
Estrogen receptors are broadly expressed across tissues, crucial for sexual development and reproductive function, and also play roles in bone and other tissues. Dysregulation is associated with breast cancer, endometrial cancer, and osteoporosis.
