The VEGF-A monoclonal antibody clone #16F1 (Product #M808) has successfully been paired as the coating antibody in a sandwich ELISA with detection antibody P802B (biotinylated conjugate of Product #P802).
Typical dilutions for sandwich ELISA include 1 µg/mL for coating and 0.125–0.5 µg/mL for detection.
Target Information
VEGF (vascular endothelial growth factor) is a 45 kDa homodimeric, disulfide-linked glycoprotein involved in angiogenesis, promoting tumor progression and metastasis. It affects vascular endothelium by enhancing cell viability, mitogenesis, chemotaxis, and vascular permeability.
The VEGF family includes VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, and PIGF. VEGF and its receptor system are key regulators of angiogenesis signaling. Most tumors require angiogenesis, and VEGF is a potent angiogenic cytokine.
Five isoforms of VEGF are produced by alternative splicing from a single VEGF gene. These isoforms bind to tyrosine-kinase receptors FLT1 (VEGFR-1) and FLK1/KDR (VEGFR-2), mainly expressed on endothelial cells. A soluble form of FLT1 (sFLT1) binds VEGF with high affinity and inhibits angiogenesis. Elevated VEGF levels are associated with POEMS syndrome (Crow-Fukase syndrome), affecting multiple organs.
