Product Specific Information

This AF6-120.1 monoclonal antibody reacts with the mouse MHC Class II I-Ab alloantigen of H-2b bearing mouse strains, including C57Bl/6 and 129. This molecule is involved in antigen presentation to T cells expressing CD3/TCR and CD4.

The antibody crossreacts with H-2k and H-2u haplotypes. Reactivity to other haplotypes (d, f, g7, p, q, r, s) has not been observed.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Optimal titration recommended.

Target Information

I-Ab (H2-Ab1, histocompatibility 2, class II antigen A, beta 1) is an MHC class II heterodimer of alpha (31–34 kDa) and beta (26–29 kDa) chains.
MHC class II molecules present exogenous antigens to CD4+ T lymphocytes, playing a key role in immune response.
They are expressed on antigen-presenting cells such as B cells, dendritic cells, and macrophages.
H2-Ab1 gene contains 6 exons encoding leader peptide, extracellular domains, transmembrane domain, and cytoplasmic tail.
Polymorphisms in alpha and beta chains determine peptide-binding specificity, relevant for immunogenetics and transplantation typing.

HLA and MHC antibodies are critical in Immunopeptidomics for neoantigen identification via LC-MS/MS.
