Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The antibody DNA sequences from immunoreactive rabbits are cloned, and individual clones are screened to select optimal candidates for production.

Target Information

Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene.
This protein is a key component of the extracellular matrix in cartilaginous tissue, providing resistance to compression. The human form (≈250 kDa, 2316 amino acids) exists in multiple isoforms via alternative splicing.

Aggrecan has a bottlebrush structure with chondroitin sulfate and keratan sulfate chains attached to its core. It contains three interglobular domains (G1, G2, G3) involved in aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis.

Aggrecan fragments are released into synovial fluid during all stages of osteoarthritis due to cleavage by aggrecanase and MMP. Antibodies BC-3 and BC-4 detect neo epitopes of Aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
