Product Specific Information

The eBio1D3 monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except plasma cells. Follicular dendritic cells also express CD19. The molecule forms a multimolecular complex with CD21, CD81, and MHC class II that associates with the BCR, inducing tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
For use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes at ≤0.2 µg/test (100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Recommended viability dyes: Invitrogen LIVE/DEAD Fixable Dead Cell stains.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining antibody cocktails with cells.
  • For single-color controls, add 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, featuring engineered nucleic acid dye structures for optimized fluorescence performance and minimal spectral spillover.

Internal testing indicates strain-dependent staining:

• Non-specific staining observed in SJL mice.
• No non-specific staining in BALB/c or C57BL/6 mice.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and certain myeloid leukemia cells. CD19 acts as a signal-amplifying coreceptor for the B cell receptor (BCR), regulating B cell activation and proliferation.
Mutations cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
