Description:
The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed on B cells at all developmental stages except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. Together with CD21, CD81, and MHC class II, CD19 forms a multimolecular complex associated with the BCR, mediating signaling that induces tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:

• Flow cytometric analysis of mouse splenocytes.

Applications Tested:

• Recommended ≤ 0.4 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).
• Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Caution:
Internal testing shows NovaFluor Yellow 700 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on all peripheral B cells and certain myeloid leukemia cells. CD19 acts as a signal-amplifying co-receptor for the B cell receptor (BCR), regulating B cell activation and proliferation. Mutations in CD19 can cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
