Product Specific Information

The eBio1D3 monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, and MHC class II, playing a key role in B cell receptor signaling, inducing tyrosine phosphorylation, calcium flux, and proliferation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes, ≤0.2 µg/test (100 µL final volume). Optimal antibody titration recommended.

Important Notes:

• NovaFluor dyes are incompatible with DNA-intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample or per 100 µL cell suspension (10³–10⁸ cells).
• NovaFluor conjugates utilize Phiton™ technology for engineered fluorescence with minimized spillover.
• Excitation: 552 nm; Emission: 690 nm; Laser: 561 nm (Yellow).
• Non-specific staining observed in SJL mice; not in BALB/c or C57BL/6.

Target Information

CD19 is a 95 kDa transmembrane glycoprotein of the immunoglobulin superfamily with two Ig-like domains. It is expressed on all peripheral B cells and certain myeloid leukemia cells. CD19 regulates B cell activation and proliferation and serves as a co-receptor with the BCR to lower activation thresholds. Mutations lead to hypogammaglobulinemia; overexpression causes B cell hyperactivity.
