Target Information

GFI1b (zinc finger protein GFI-1b) is an essential proto-oncogenic transcriptional regulator necessary for development and differentiation of erythroid and megakaryocytic lineages. It is part of the RCOR-GFI-KDM1A-HDAC complex that suppresses genes involved in multilineage blood cell development through histone deacetylase (HDAC) recruitment.
GFI1b acts as a transcriptional repressor or activator depending on promoter and cell type context, repressing SOCS1 and SOCS3 promoter activity to regulate cytokine signaling pathways. It cooperates with GATA1 to repress target gene transcription such as BCL2L1, and silencing of GFI1B in leukemic cell lines markedly increases apoptosis rate.
Mutations in GFI1B are associated with Bleeding disorder, platelet-type 17 (BDPLT17).
